平常的人一生中初恋仅仅有一次，但亨利·罗斯(亚当·桑德勒饰)却遇上了50次初恋，而且都是跟同一个女孩，或许你会为他感叹，但当你看到和亨利“初恋”的女孩露茜时，你就肯定会嫉妒得要死了。亨利·罗斯是夏威夷水族馆的一名兽医，也是一位快乐的单身汉，在管理海洋动物的同时，他还是这个领域的科学家。单身的亨利是

个憨厚老实的人，他一直与他的海洋动物们过着快乐的生活，但有一次在咖啡馆的邂逅，改变了他的平静生活。

那天，亨利下班后走进了水族馆旁边的咖啡馆，他和朋友想吃点东西。正当他们坐在那里等食物的时候，一道镜子的反射闪光照到了亨利的脸上，亨利自然地回过头来，天呀！他看到了一位金发公主，那女孩有着如火般的金发，圆润的脸上洋溢出快乐，她的身材丰满娇媚。亨利怦然心动，这不就是自己一直寻找的梦中情人吗？

经过多方打听，亨利终于了解到，露茜因为一次车祸而患上了一种极为罕见的短期记忆丧失症。当第二天太阳升起的时候，她就会把前一天的事情全都忘光。

坠入情网的亨利无法放弃自己心爱的女孩，为了自己，也为了向女孩的父亲证明自己对她的爱。为了与梦中情人露茜保持恋人关系，亨利在朋友尤拉(罗伯·施奈德饰)的帮助下，开始了既疯狂又令人感动的追求计划——务求在太阳升起，每一个新的一天来临之时，让露茜再一次爱上他。在这50次初恋中，发生了许许多多令人捧腹大笑又温馨的故事。
